Scanning electron microscopy (SEM) requires vacuum conditions and therefore all samples need to be images in dry state. Particularly for biological samples, the final dehydration step is typically done using critical point drying. Following complete dehydration, non-conductive specimens are sputter coated with a thin conductive layer to prevent charging during electron microscopy. We have two automated sputter coaters for gold and carbon coatings.
