  This study investigates the effect of scaffold architecture on bone regeneration, focusing on 3D-printed polylactic acid-bioceramic calcium phosphate (PLA-bioCaP) composite scaffolds in rabbit femoral condyle critical defects. We explored two distinct scaffold designs to assess their influence on bone healing and scaffold performance. Structures with alternate (0° /90°) and helical (0°/45°/90°/135°/180°) laydown patterns were manufactured with a 3D printer using a fused deposition modeling technique. The scaffolds were meticulously characterized for pore size, strut thickness, porosity, pore accessibility, and mechanical properties. The in vivo efficacy of these scaffolds was evaluated using a femoral condyle critical defect model in eight skeletally mature New Zealand White rabbits. Then, the results were analyzed micro-tomographically, histologically, and histomorphometrically. Our findings indicate that both scaffold architectures are biocompatible and support bone formation. The helical scaffolds, characterized by larger pore sizes and higher porosity, demonstrated significantly greater bone regeneration than the alternate structures. However, their lower mechanical strength presented limitations for use in load-bearing sites.

  Introduction: The effectiveness of Multiple-Choice Questions (MCQs) in dental education is pivotal to student performance and knowledge advancement. However, their optimal implementation requires exploration to enhance the benefits. Materials and Methods: An educational tool incorporating MCQs was administered from the 5th to the 10th semester in a dental curriculum. The students filled out a questionnaire after the MCQ, which was linked to the Learning Management System. 4 cohorts of 4 semesters generated 2300 data points analysed by Spearmen correlation and mixed model regression analysis. Results demonstrated a significant correlation between early exam preparation and improved student performance. Independent study hours and lecture attendance emerged as significant predictors, accounting for approximately 10.27% of the variance in student performance on MCQs. While the number of MCQs taken showed an inverse relationship with study hours, the perceived clarity of these questions positively correlated with academic achievement. Conclusion: MCQs have proven effective in enhancing student learning and knowledge within the discipline. Our analysis underscores the important role of independent study and consistent lecture attendance in positively influencing MCQ scores. The study provides valuable insights into using MCQs as a practical tool for dental student learning. Moreover, the clarity of assessment tools, such as MCQs, remains pivotal in influencing student outcomes. This study underscores the multifaceted nature of learning experiences in dental education and the importance of bridging the gap between student expectations and actual performance.

  The vascular tree is crucial for the survival and function of large living tissues. Despite breakthroughs in 3D bioprinting to endow engineered tissues with large blood vessels, there is currently no approach to engineer high-density capillary networks into living tissues in a scalable manner. We here present photo-annealing of living microtissues(PALM) as a scalable strategy to engineer capillary-rich tissues. Specifically, in-air microfluidics was used to produce living microtissues composed of cell-laden microgels in ultra-high throughput, which could be photo-annealed into a monolithic living matter. Annealed microtissues inherently give rise to an open and interconnected pore network within the resulting living matter. Interestingly, utilizing soft microgels enables microgel deformation, which leads to the uniform formation of capillary-sized pores. Importantly, the ultra-high throughput nature underlying the microtissue formation uniquely facilitates scalable production of living tissues of clinically relevant sizes (>1 cm3 ) with an integrated high-density capillary network. In short, PALM generates monolithic, microporous, modular tissues that meet the previously unsolved need for large engineered tissues containing high-density vascular networks, which is anticipated to advance the fields of engineered organs, regenerative medicine, and drug screening

  Objectives: This study aimed at identifying biomarkers in the temporomandibular joint (TMJ) synovial tissue analysing 28 extra cellular matrix proteins in TMJ diseased patients, classified with either general joint hypermobility (GJH) or normal joint mobility (NJM), and to compile clinical and protein characterisation to reveal potential surgical predictive factors. Study design: A prospective observational cohort study including 97 consecutive patients scheduled for TMJ surgery was performed. Joint mobility and several other predefined clinical varivables were recorded. Synovial tissue was harvested during surgery followed by examination using multi-analytic profiling. A multivariate quantile regression model was used for analysis purposes. Results: The GJH/NJM ratio was 2:5. The GJH cohort were younger (P = 0.001) and more likely to be women (P = 0.026) compared to the NJM cohort. None of the protein concentrations could be correlated to joint mobility in the multivariate regression model, but often to the variable TMJ diagnosis. The surgical outcome after the six-month follow-up were equal between GJH and NJM patients. Conclusions: GJH was more common in the study cohort compared to general population frequencies, but GJH was not a negative factor for surgical outcome. Young age and female gender correlated to GJH. No TMJ biomarkers were GJH specific, and the results suggested that TMJ diagnosis more strongly correlated to the protein profile compared to GJH and the other investigated variables.

  Objectives: This study aimed to determine the degree of similarity and symmetry in the anatomy of contralateral mandibular incisors. Three-dimensional (3D) models of extracted teeth were obtained from microtomography (micro-CT) scans. Qualitative and quantitative assessments of the morphology and comparison of contralateral pairs were made. The null hypothesis was that contralateral mandibular incisors could not be considered identical in simple morphometric measurements. Methods: Sixty pairs of mandibular incisors were extracted from 30 patients and scanned with micro-CT. Virtual models of the cemento-enamel junction to the root apex were rendered. Parameters such as length, canal width, dentinal thicknesses, tortuosity, centerline length, accessory canals, root canal configurations, and root canal orifice cross-sections were used to compare the teeth. Width and thickness comparisons between paired teeth in the same individual were made by paired t-test (Wilcoxon Signed Rank Test for variables not normally distributed). An online randomization tool generated randomized pairs (independent of the individual/patient). Subsequently, an unpaired t-test (or Mann-Whitney U test for non-normally distributed parameters) and a correlation analysis were conducted. Canal configurations were classified according to preexisting classification schemes. The number and location of accessory canals and apical foramina were registered and compared. Results: Utilizing advanced imaging techniques and quantitative analyses, our study establishes that contralateral mandibular incisors exhibit a remarkable degree of symmetry in multiple morphological parameters, including length, canal width, and dentinal thicknesses The apical third showed a high degree of inter-variability for the contralateral pairs. The rigorous statistical analysis of the normalized parameters by Z score showed no statistically significant differences between the contralateral mandibular incisors. Comparisons between central and lateral teeth revealed differences in root length but no significant disparity in the distribution of accessory canals. Central teeth, on average, were longer, while accessory canals were distributed relatively evenly between central and lateral teeth. Conclusions: The findings of this study further establish the significant similarities between contralateral mandibular incisors, reinforcing their suitability as a reliable substrate for root canal comparison studies. Clinical Relevance: The absence of statistically significant differences between contralateral pairs in normalized parameters underscores their potential as a reliable reference point for root canal comparison studies in clinical dentistry. Furthermore, our findings emphasize the importance of individualized treatment planning, considering the natural symmetry in mandibular incisors to enhance clinical decision-making. This research contributes valuable insights to the field of endodontics, offering a standardized approach to sample selection and enriching the understanding of dental anatomy

  Ameloblastin (AMBN) is best characterized for its role in dental enamel formation, regulating cell differentiation and mineralization, and cell matrix adhesion. However, AMBN has also been detected in mesenchymal stem cells in addition to bone, blood, and adipose tissue. Using immunofluorescence in a pilot scheme, we identified that AMBN is expressed in different parts of the gastrointestinal (GI) tract. AMBN mRNA and protein detection in several tissues along the length of the GI tract suggests a role for AMBN in the structure and tissue integrity of the extracellular matrix (ECM). Intracellular AMBN expression in subsets of cells indicates a potential alternative role in signaling processes. Of note, our previous functional AMBN promoter analyses had shown that it contains epithelial–mesenchymal transition (EMT) regulatory elements. ΑΜΒΝ is herein presented as a paradigm shift of the possible associations and the spatiotemporal regulation of the ECM regulating the EMT and vice versa, using the example of AMBN expression beyond oral biology.

  Micro-computed tomography (micro-CT) provides valuable data for studying soft tissue, though it is often affected by sample movement during scans and low contrast in X-ray absorption. This can result in lower image quality and geometric inaccuracies, collectively known as 'artefacts'. To mitigate these issues, samples can be embedded in hydrogels and enriched with heavy metals for contrast enhancement. However, the long-term durability of these enhancements remains largely unexplored. In this study, we examine the effects of two contrast enhancement agents – iodine and phosphotungstic acid (PTA) – and two hydrogels – agarose and Poloxamer 407 – over a 14-day period. We used Drosophila melanogaster as a test model for our investigation. Our findings reveal that PTA and agarose are highly durable, while iodine and poloxamer hydrogel exhibits higher leakage rates. These observations lay the foundation for estimating contrast stabilities in contrast-enhanced micro-CT with hydrogel embedding and serve to inform future research in this field.

  The role of poroelasticity on the functional performance of articular cartilage has been established in the scientific literature since the 1960s. Despite the extensive knowledge on this topic there remain few attempts to design for poroelasticity and to our knowledge no demonstration of an engineered poroelastic material that approaches the physiological performance. In this paper, we report on the development of an engineered material that begins to approach physiological poroelasticity. We quantify poroelasticity using the fluid load fraction, apply mixture theory to model the material system, and determine cytocompatibility using primary human mesenchymal stem cells. The design approach is based on a fiber reinforced hydrated network and uses routine fabrication methods (electrohydrodynamic deposition) and materials (poly[ɛ-caprolactone] and gelatin) to develop the engineered poroelastic material. This composite material achieved a mean peak fluid load fraction of 68%, displayed consistency with mixture theory, and demonstrated cytocompatibility. This work creates a foundation for designing poroelastic cartilage implants and developing scaffold systems to study chondrocyte mechanobiology and tissue engineering.

  In 1892, J.L. Wolff proposed that bone could respond to mechanical and biophysical stimuli as a dynamic organ. This theory presents a unique opportunity for investigations on bone and its potential to aid in tissue repair. Routine activities such as exercise or machinery application can exert mechanical loads on bone. Previous research has demonstrated that mechanical loading can affect the differentiation and development of mesenchymal tissue. However, the extent to which mechanical stimulation can help repair or generate bone tissue and the related mechanisms remain unclear. Four key cell types in bone tissue, including osteoblasts, osteoclasts, bone lining cells, and osteocytes, play critical roles in responding to mechanical stimuli, while other cell lineages such as myocytes, platelets, fibroblasts, endothelial cells, and chondrocytes also exhibit mechanosensitivity. Mechanical loading can regulate the biological functions of bone tissue through the mechanosensor of bone cells intraosseously, making it a potential target for fracture healing and bone regeneration. This review aims to clarify these issues and explain bone remodeling, structure dynamics, and mechano-transduction processes in response to mechanical loading. Loading of different magnitudes, frequencies, and types, such as dynamic versus static loads, are analyzed to determine the effects of mechanical stimulation on bone tissue structure and cellular function. Finally, the importance of vascularization in nutrient supply for bone healing and regeneration was further discussed

  Abstract Background Irisin is expressed in human periodontal ligament (hPDL), and its administration enhances growth, migration and matrix deposition in hPDL cells cultured in monolayers in vitro. Objectives To identify whether irisin affects the gene expression patterns directing the morphology, mechanical properties, extracellular matrix (ECM) formation, osteogenic activity and angiogenic potential in hPDL cell spheroids cultured in 3D. Materials and Methods Spheroids of primary human hPDL cells were generated in a rotational 3D culture system and treated with or without irisin. The gene expression patterns were evaluated by Affymetrix microarrays. The morphology of the spheroids was characterized using histological staining. Mechanical properties were quantified by nanoindentation. The osteogenic and angiogenic potential of spheroids were assessed through immunofluorescence staining for collagen type I, periostin fibronectin and von Willebrand factor (vWF), and mRNA expression of osteogenic markers. The secretion of multiple myokines was evaluated using Luminex immunoassays. Results Approximately 1000 genes were differentially expressed between control and irisin-treated groups by Affymetrix. Several genes related to ECM organization were differentially expressed, and multiple deubiquitinating enzymes were upregulated in the irisin-exposed samples analyzed. These represent cellular and molecular mechanisms indicative of a role for irisin in tissue remodeling. Irisin induced a rim-like structure on the outer region of the hPDL spheroids, ECM-related protein expression and the stiffness of the spheroids were enhanced by irisin. The expression of osteogenic and angiogenetic markers was increased by irisin. Conclusions Irisin altered the morphology in primary hPDL cell-derived spheroids, enhanced its ECM deposition, mechanical properties, differentiation and remodeling potential.

  Background Microgravity directly disturbs the reorganization of the cytoskeleton, exerting profound effects on the physiological process of macrophages. Although it has been established that macrophage M1/M2 polarization could be manipulated by the surface nanostructure of biomaterial in our previous study under normal gravity, how will inflammatory monocytes (iMos)-derived macrophages respond to diverse nanostructured Ti surfaces under normal gravity or microgravity remains unrevealed. Results In this study, Cytochalasin D, a cytoskeleton relaxant, was employed to establish the simulated microgravity (SMG) environment. Our results showed that human iMos polarized into M2c macrophages on NT5 surface but M1 type on NT20 surface with divergent inflammatory phenotypes according to the profile of macrophage polarization featured molecules under normal gravity. However, such manipulative effects of NTs surfaces on iMos-derived macrophages were strikingly weakened by SMG, characterized by the altered macrophage morphology, changed cytokine secretion profile, and decreased cell polarization capacity. Conclusions To our knowledge, this is the first metallic implantable material study focusing on the functions of specific monocyte subsets and its crucial role of the cytoskeleton in materials-mediated host immune response, which enriches our mechanism knowledge about the crosstalk between immunocytes and biomaterials. The results obtained in the present study may also provide potential targets and strategies for biomaterial development and clinical treatment via precise immune-regulation under normal gravity and microgravity.

  Biphasic monolithic materials for the treatment of osteochondral defects were produced from polysaccharide hydrogel, gellan gum (GG). GG was enzymatically mineralized by alkaline phosphatase (ALP) in the presence of calcium glycerophosphate (CaGP). The desired distribution of the calcium phosphate (CaP) mineral phase was achieved by limiting the availability of CaGP to specific parts of the GG sample. Therefore, mineralization of GG was facilitated by the diffusion of CaGP, causing the formation of the CaP gradient. The distribution of CaP was analyzed along the cross section of the GG. The formation of a CaP gradient was mainly affected by the mineralization time and the ALP concentration. The formation of CaP was confirmed by Fourier transform infrared spectroscopy (FTIR), Raman spectroscopy and mapping, as well as energy-dispersive X-ray spectroscopy (EDX) mapping of the interphase. The microstructure of mineralized and non-mineralized parts of the material was characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM) showing sub-micrometer CaP crystal formation, resulting in increased surface roughness. Compression tests and rheometric analyzes showed a 10-fold increase in stiffness of GG mineralized part. Concomitantly, micromechanical tests performed by AFM showed an increase of Young's modulus from 17.8 to more than 200 kPa. In vitro evaluation of biphasic scaffolds was performed in contact with osteoblast-like MG-63 cells. The mineralized parts of GG were preferentially colonized by the cells over the non-mineralized parts. The results showed that osteochondral scaffolds of the desired structure and properties can be made from GG using a diffusion-limited enzymatic mineralization method.

  Gastro oesophagael reflux disease (GORD) is common in the Western hemisphere. Patients with regurgitated reflux are typically treated with fundoplication surgery. We present a newly designed polyurethane implant which passively aids the sphincter in reducing gastric fluids within the oesophagus. The gastric implant has an open porous inner side which allows for tissue ingrowth from the oesophagus and thus allows for fixation around the sphincter. In addition, a device for minimally invasive surgery of this implant was developed and used in a pig model. The unmodified GORD implant was placed around the pig’s oesophagus with unsatisfactory results, leading to insufficient fixation at the implantation site and scarring tissue leading to dysphagia. In addition, two surface modifications, plasma activation and TiO2 deposition were used to improve the implant’s host tissue response. The biocompatibility effects of the surface treatments and sterilisation method on the implant were investigated in vitro and in vivo. In vitro tests found that the plasma activation and TiO2 deposition have effectively enhanced the surface hydrophilicity and, consequently, the cell response to the implant. In addition, the gamma sterilisation harmed the plasma-activated implant. The plasma activation was more effective than TiO 2 deposition as a surface treatment method for improving the tissue response of this implant in vivo. In addition, the in vivo experiment proved tissue ingrowth as deep as 1 mm into the porous structure of the implant. The GORD implants were encapsulated wholly in fibrous tissue; however, the capsule thickness diminished over time. Finally, the TiO2-coated implants showed the poorest histocompatibility, contradictory to the in vitro findings. This study shows that it is possible to produce a plasma-treated porous polyurethane gastric implant that allows for fibrous tissue ingrowth, reduced in vivo encapsulation, and enhanced chemical properties

  Hydrogels based on hyaluronic acid (HA) and agarose–carbomer (AC) units have been prepared and explored as drug delivery systems. The complex architecture of the polymer network, such as mesh size, HA molecular weight and drug-polymer non-covalent interactions across the 3D polymer matrix, strongly influence the release capability/profile of these materials. In this study, High-Resolution Magic Angle Spinning (HR-MAS) NMR Spectroscopy has been used to investigate the transport behaviour of two different drugs, such as ethosuximide (neutral molecule) and sodium salicylate (net negative charge) within the AC and AC-HA hydrogel networks prepared with different mesh sizes. Analysis of the experimental data provides evidence of superdiffusive motion for all formulations containing sodium salicylate, while ethosuximide molecules undergo unrestricted diffusion within the gel matrix. We further speculate that the superdiffusive motion observed at the nanoscale can be responsible for the faster release of sodium salicylate from all hydrogel formulations.

  Nanotechnology has become an emerging research field with numerous biomedical scientific applications. Silver possesses bactericidal activities that have been harnessed for centuries; however, there is a concern about the toxic effects of silver nanoparticles. This paper aims to provide an overview of silver-treated dental implants and discuss their potential to reduce the prevalence of peri-implant diseases. An electronic search was performed using PubMed. After screening, data extraction was performed on the 45 remaining articles using inclusion and exclusion criteria. Most of the articles demonstrated that silver nanoparticles embedded in a coating layer and/or on surfacetreated titanium exhibit sound antibacterial effects and biocompatibility. Most of the reviewed studies revealed that silver nanoparticles on dental implant surfaces reduced cytotoxicity but provided a prolonged antibacterial effect. The cytotoxicity and antibacterial effect are closely linked to how the silver nanoparticles are released from the titanium surfaces, where a slower release increases cell viability and proliferation. However, to improve the clinical translation, there is still a need for more studies, especially evaluating the long-term systemic effects and studies recreating the conditions in the oral cavity.

  Bone grafts are considered to be the gold standard in terms of repairing and reconstructing damaged bones. These can either be autologous, where a patient’s own tissue is used, or allogeneic, where the tissue is taken from another person. But limited supply, donor-site complications, and risk of disease transmission can often prevent allogeneic grafts from being used. Xenogeneic bone grafts, where the graft is taken from another species, represent a feasible alternative. But, because xenograft materials come from mammal tissue, it raises ethical questions surrounding animal welfare. To find a safe, adaptable, and environmentally friendly alternative, a team of researchers has developed a method to convert eggshells into endotoxin-free and immunocompatible amorphous calcium phosphate (ACP) particles. First the shells were heated, then mixed with distilled water and phosphoric acid. The precipitate was then filtered and washed, before being submerged in liquid nitrogen. The resulting ACP particles are essential to forming hard and strong bone and can therefore be used as an ideal bone substitute. As well as containing plenty of calcium and phosphorous, Dr Qianli Ma – the study’s lead author – highlights that eggshells are also an ideal raw material because they contain traces of magnesium and strontium, which are associated with healthy bone regeneration. The team also created a novel 3D spheroid model, which allows the activity of eggshell ACP to be studied in vitro. With the model, the researchers could observe how the ACP materials would actually interact with osteoblasts, and they were found to be safe and effective in promoting bone regeneration. If scaled successfully, this new technique could enable an unlimited supply of sustainable bone graft materials, while reducing the volume of eggshells going to waste. The scientists hope their latest findings inspire additional research into the conversion of food waste into high-value biomaterials. Eggshells are not only being used in the medical field. Springwise has also spotted a design studio creating wall tiles from discarded eggshells, and another that has transformed the waste into car interiors
